摘要:
建立了悬浮溶剂固化-分散液液微萃取法提取小米中β-胡萝卜素的方法,并用紫外-可见分光光度计测定其含量.结果表明,最佳提取条件是:取4 mL小米样品的皂化液,用5 mL蒸馏水稀释到10 mL离心管中,调节pH值为7、加入NaCl至饱和,快速加入混匀的200 μL十二醇和200 μL乙腈溶液萃取,完成后将离心管迅速置于冰水浴中 静置5 min,用微量注射器吸出残余样品液后,室温融化固化的萃取剂用四氢呋喃溶解并定容至1 000 μL.萃取完成后将提取液用紫外-可见分光光度计在波长460 nm处测定小米中β-胡萝卜素含量.该方法工艺简单,提取率高.
Abstract:
A novel method for the extraction and determination of β-carotene in millet has been developed by dispersive liquid-liquid microextraction combined with solidified floating organic droplets(SFO-DLLME)and ultraviolet visible spectrophotometer.The results show that the optimal parameters are diluting 4 mL of saponified solution with 5 mL water,adjusting pH=7,adding saturated NaCl and 200 μL 1-dodecanol(extractant)with the same volume of acetonitrile(dispersant)to the aqueous solution for extracting.After that,the centrifuge tube is immersed in an ice-water bath for 5 min,the floated organic solvent is solidified on top of the surface of the aqueous solution after a short period of time,and any liquid phase is removed using the same syringe.The solidified floating is quickly melted at room temperature and the extract is diluted to 1 000 μL using THF.Ultimately,β-carotene in the extract is determined by ultraviolet visible spectrophotometer.The advantage of the extraction method shows simplicity of the procedure,high extraction rate,which is available for the extraction and determination of β-carotene in millet.
